    血清胸腺因子(FTS)的合成和活性测定

    Synthesis and Activity Determination of Thymulin(FTS)

    • 摘要: 采用芴甲氧羰基(Fmoc)固相肽合成法(Fmoc~SPPS)合成了血清胸腺因子(FTS),用中压液相阴离子柱一步纯化,经HPLC分析纯度,通过质谱和氨基酸序列测定鉴定合成产物。根据FTS恢复胸腺切除小鼠的脾细胞对免疫抑制荆AZ敏感的特性测定合成FTS的活性。结果表明:化学合成血清胸腺因子的产率为94.7%;纯化后其纯度达到93.7%;质谱测定其相对分子质量为876.6,与理论分子量相符。氨基酸序列测定为NH2/Glu—Ala—Lys—Ser—Gin—Gly—Gly—Ser—Asn,与设计序列一致。实验显示在合成FTS的浓度为10^-14mol/L时尚有部分活力,在10^-13mol/L时能够完全恢复胸腺切除小鼠的脾细胞对AZ的敏感性。

       

      Abstract: Thymulin(FTS) was synthesized by Fmoc-SPPS,purified by MPLC,and identified by HPLC,MS and amino acid sequence determination.Biological activity of FTS was measured according to its characteristic of conferring on spleen cells from adult thymectomized that mice sensitivity to azathioprine(AZ) identical to that of normal mouse spleen cells.Results show the purity of the product was (93.7%);the molecular mass was 876.6;the amino acid sequence was NH_2/Glu-Ala-Lys-SerGln-Gly-Gly-Ser-Asn.The synthesized FTS can completely restore spleen cells from thymectomized mice sensitivity to AZ in the concentration of 10~(-13) mol/L.
