    低氧诱导启动子PsADH2在毕赤酵母中的调控特性

    Regulation Characteristics of Hypoxia Induced PsADH2 Promoter in Pichia pastoris

    • 摘要: 树干毕赤酵母(Pichia stipitis)乙醇脱氢酶Ⅱ(Alcohol dehydrogenase Ⅱ)基因PsADH2的启动子是低氧诱导型启动子。利用透明颤藻血红蛋白(VHb)基因vgb、绿色荧光蛋白(GFP)基因gfp和β 半乳糖苷酶(Gal)基因lacZ作为报告基因,从转录和蛋白水平初步探讨了PsADH2启动子在毕赤酵母中的调控特性。结果表明:3个报告基因在PsADH2启动子的调控下,转录和蛋白水平均随溶氧的降低而升高,报告基因的表达对菌体生长无影响,这为构建新的诱导表达系统提供了有益的数据。

       

      Abstract: The PsADH2 promoter of Pichia stipitis alcohol dehydrogenase Ⅱ (ADH Ⅱ) gene has been shown to be induced by oxygen limitation. vgb, gfp and lacZ was used as report genes to study the regulation of PsADH2 promoter in Pichia pastoris from mRNA and protein level. Results indicated that with the decreasing of dissolved oxygen, the expression of report genes was improved in both mRNA and protein level, and the expression of heterologous genes didn’t affect cell growth, which provided useful data for constructing new induced expression systems.
