    基于AHP-CRITIC赋权结合响应面法多指标优选醉鱼草的微波提取工艺

    Microwave Extraction Process for Multi-Index Components of Buddleja lindleyana Fortune Optimized Based on AHP-CRITIC Weighting Combined with Response Surface Methodology

    • 摘要: 以蒙花苷、毛蕊花糖苷、金合欢素得率及出膏率为评价指标,通过单因素实验,考察粒度、微波功率、液固比、提取溶剂、提取时间以及提取次数对醉鱼草提取效果的影响;应用Box-Behnken响应面法设计实验,结合层次分析-批判(AHP-CRITIC)组合赋权法对各评价指标进行主客观赋权,以综合得分为响应值,优化醉鱼草微波提取工艺并进行体外抗氧化活性研究。结果表明:最优提取条件为微波功率325 W,液固比(提取溶剂的体积与药材质量之比)65 mL/g,乙醇质量分数66%,提取时间8 min,在此条件下,蒙花苷、毛蕊花糖苷、金合欢素的得率分别为5.417%、0.728%和0.175%,出膏率为25.769%,综合评分为99.19(相对标准偏差0.45%)。优化后的提取工艺操作简便、稳定性好,且所得提取物具有较好的抗氧化活性。

       

      Abstract: The effects of particle size, microwave power, liquid-solid ratio, extraction solvent, extraction time, and extraction times on the microwave extraction of Buddleja lindleyana Fortune, using the yields of linarin, acteoside, acacetin and the paste as evaluation indexes, were investigated through single-factor experiments. The analytic hierarchy process (AHP) and criteria importance through inter-criteria correlation (CRITIC) method were combined to conduct an objective and subjective weighting of each evaluation index, and the Box-Behnken response surface methodology (RSM) was applied to design a total of 29 experiments with four factors at three levels. With the comprehensive score serving as the response value, the microwave extraction process for Buddleja lindleyana Fortune was optimized and the in vitro antioxidant activity of the extract was tested. With a fixed particle size of 120-mesh (120 μm) and two extraction cycles, the optimal extraction conditions were obtained as follows: A microwave power of 325 W, a liquid-solid ratio of 65 mL/g, an ethanol volume fraction of 66%, and an extraction time of 8 min. With these parameters set, the yield of linarin, acteoside and acacetin was determined to be 5.417%, 0.728% and 0.175%, respectively, and the paste yield was determined to be 25.769%, with a comprehensive score of 99.19 (RSD=0.45%). The optimized extraction process is simple, stable and efficient, and the obtained extract exhibited antioxidant activity.
