    (6,5)单壁碳纳米管对盐酸阿霉素的载药研究

    Drug Delivery System Based on (6,5) Single-Walled Carbon Nanotube Loaded with Doxorubicin Hydrochloride

    • 摘要: 以分离后的单壁碳纳米管为载药工具,制备了一种(6,5)单壁碳纳米管-盐酸阿霉素-人血清白蛋白-叶酸((6,5)SWCNT-Dox-HSA-FA)四元靶向载药系统。首先通过反应制备得到三元复合物Dox-HSA-FA,并采用双水相萃取法分离得到(6,5)SWCNT;然后采用超声作用将(6,5)SWCNT分散至Dox-HSA-FA中制备得到(6,5)SWCNT-Dox-HSA-FA;最后采用Hep G2细胞对四元复合物进行细胞毒性评价。结果表明,Dox-HSA-FA和(6,5)SWCNT-Dox-HSA-FA的半数抑制浓度(IC50)分别为27.3 μg/mL和25.7 μg/mL,表明四元复合物可以提高盐酸阿霉素的靶向性。

       

      Abstract: A drug delivery system was designed in this paper. The system was composed of (6,5) chiral single-walled carbon nanotube ((6,5)SWCNT), doxorubicin hydrochloride (Dox), human serum albumin (HSA), and folic acid (FA). Firstly, the Dox-HSA-FA was prepared and (6,5) SWCNT was isolated by aqueous two-phase extraction. Then, (6,5) SWCNT-Dox-HSA-FA was dispersed into Dox-HSA-FA by ultrasonication. Finally, cytotoxicity evaluation of the complex was performed using Hep G2 cells. The results showed that the IC50 of Dox-HSA-FA and (6,5) SWCNT-Dox-HSA-FA was 27.3 μg/mL and 25.7 μg/mL, respectively. The results indicated that the targeting effect of Dox was enhanced in the (6,5) SWCNT-Dox-HSA-FA complex.
