Abstract:
Application of enzymatic pretreatment and microwave extraction (EPME) to the extraction of asiaticoside from Centella asiatica was investigated in this paper. Temperature of enzymatic pretreatment, liquid to solid ratio, microwave radiation time were examined and selected as the factors for the quadric regression orthogonal experiments to find out the optimum operating conditions for the EPME. An extraction model with well forecast performance was also established. Results of orthogonal experiments indicated that the optimum operating conditions were as follows: temperature of enzymatic pretreatment was 45 ℃, liquid to solid ratio was 36, enzymatic time was 30 min and microwave radiation time was 110 s. Under these condition the maximum yield could reach 27.10%. Compared to the traditional extraction methods, EPME was high in extracting rate and objective constituents yield. It may be considered as an ideal means for extracting asiaticoside from Centella asiatica.
