    鱼类致病性迟钝爱德华氏菌胶体金快速检测试纸的研制

    Gold Immunochromatography Dipstick for Rapid Detection of Fish Pathogen E. tarda

    • 摘要: 制备迟钝爱德华氏菌胶体金快速检测试纸并对其性能进行检测。以柠檬酸三钠作为还原剂制备胶体金颗粒,标记迟钝爱德华氏菌多克隆抗体,将羊抗兔IgG和纯化后的迟钝爱德华氏菌多克隆抗体包被于NC膜上作为质控线和检测线,以检测样品中的迟钝爱德华氏菌(E. tarda)。结果表明:所研制的试纸特异性良好,对大肠杆菌、金黄色葡萄球菌、鳗弧菌、哈维氏弧菌、嗜水气单胞菌等常见细菌或病原菌进行测试未出现交叉反应;敏感度达到105 CFU/mL,反应时间仅为5~10 min,可以用于感染迟钝爱德华氏菌大菱鲆腹水的现场检测,具有操作

       

      Abstract: The research aims to develop colloidal gold immunochromatographic test strip for rapid detection of E. tarda. The purified polyclonal antibody was labeled with the colloidal gold which was synthesized by trisodium citrate disoxidation method. Goat anti-rabbit IgG and purified polyclonal antibody were separately sprayed on the nitrocellulose membrane as the controlline (C line) and the test line (T line) to detect E. tarda in the samples. The results indicated that E. tarda gold immunochroamatography diagnostic strips established in this study displayed perfect specificity without cross-reactions with E. coli, S. aureus, V. anguillarum, V. harveyi, A. hydrophilia and E. ictaluri. Besides the high specificity, its sensitivity reached 105 CFU/mL and the reaction time only need 5 to 10 minutes. The dipstick was also used for detection of E. tarda in infected turbot ascites fluid. The results revealed that the test strip has good sensitivity, specificity and stability, which is easy to operate and can be used for rapid diagnosis of E. tarda.
