    非修饰性寡核苷酸芯片用于HLA-DQA分型的初步研究

    Preliminary Study on HLA-DQA Genotyping by Unmodified-Oligonucleotide Microarrays

    • 摘要: 利用非修饰寡核苷酸基因芯片技术,根据HLAⅡ类抗原DQA位点不同基因亚型的特异性序列设计探针,制成分型芯片;待测样本经PCR反应标记上荧光之后,与探针在芯片上进行杂交,根据杂交产生的荧光信号值,确定样品DQA位点基因亚型。将这一方法应用于125例临床样本和10例标准品的HLA-DQA分型,该方法分型的结果与准确结果相比,准确率达到93.3%。实验结果表明:非修饰性寡核苷酸芯片用于HLA-DQA分型技术可行。

       

      Abstract: According to the sequence of HLA-DQA exon2,HLA-DQA genotyping microarray was made and the labeled PCR products were hybridized with arrays.The signals were scanned by scanner and analyzed by image software.We genotyped 125 clinical samples and 10 reference samples.The accuracy of identifying the HLA-DQA genotyping using unmodified-oligonucleotide microarray was 93.3% compared with the actual results.Unmodified-oligonucleotide microarray provides a high-specific,inexpensive,(reproducible,) rapid and intuitional tool for HLA genotyping and serological typing could be done with such microarray in routine medical practice.
