    Enzymatic Synthesis of 5 Methyluridine by Brevibacterium acetylicum

    • 摘要: 用乙酰短杆菌完整细胞为酶源,以鸟苷和胸腺嘧啶为底物,酶法合成5-甲基尿苷。最佳反应条件为:底物浓度100-200mmol/L,pH8.0,磷酸盐缓冲液尝试10-50mmol/L,60℃,摇床转速120r/min,反应1h即达到平衡,5-甲基尿苷转化率达60%以上。

       

      Abstract: Methyluridine was enzymatically produced from guanosine and thymine by intact cells of Brevibacterium acetylicum QD9805. The optimal reaction conditions were researched. When the reaction mixtures, containing 100200mmol/L substrates, 1050mmol/L phosphate buffer(pH 8.0),were incubated at 60C shaking at 120r/min for 1h, the conversion yield could reach above 60%.
