    Effect of the Seeding Density on the Growth of Vero Cells on Microcarriers

    • 摘要: 研究了接种密度对Vero细胞在微载体表面生长行为的影响,发现培养过程中所获得的细胞最高密度随接种密度的增加而有所提高,当细胞的表面接种密度高于1.0×10^5cells/mgMC时,因贴壁后细胞扩展受到限制而不利于进入对数生长期。就整个Vero细胞培养过程,适宜的表面接种密度为3×10^-4-7×10^4cells/mgMC,此时可以获得较高的比生长速率和细胞增殖倍数。

       

      Abstract: The effect of cell seeding density on the growth of Vero cells on microcarriers was investigated. It was found that the maximal cell density obtained in the culture process increased with increasing inoculum size. However, if the surface seeding density higher than 1.0105 cells/mgMC, the cell growth was hindered to enter the exponential growth phase, due to the limitation of cell extension after attachment. The experimental results showed that the suitable cell seeding density on microcarrier surfaces was in the range of 3104 to 7104 cells/mgMC, in which the specific growth rate and the cell expansion ratio would reach high. In addition, it was indicated that the maximal cell density on microcarriers CT3 as monolayers was about 5.2105 cells/mg MC.
