    Inhibition of Diosgenin and Its Analogues Transformed by Streptomyces virginiae IBL-14 on Human Breast Cancer Cells (Bcap37) in Vitro

    • 摘要: 通过维吉尼链霉菌(Streptomyces virginiae) IBL-14对薯蓣皂苷元进行生物转化,得到一系列新的代谢产物;用四唑蓝(MTT)比色法测定薯蓣皂苷元及其衍生物对Hela、Bcap37、KB-3-1、SW1990 4种肿瘤细胞的体外抑制作用;用Hoechst 33258细胞核荧光染色来观察薯蓣皂苷元及其衍生物对Bcap37细胞形态学上的改变;采用流式细胞计量法测定薯蓣皂苷元及其衍生物对Bcap37细胞的凋亡作用。结果表明:薯蓣皂苷元及其3个衍生物在一定浓度范围内,对上述4种肿瘤细胞均有抑制生长的作用,其中对人乳腺癌细胞 Bcap37的抑制效果相对显著。在薯蓣皂苷元及其衍生物的影响下,人乳腺癌细胞Bcap37细胞呈现典型的细胞凋亡特征,包括细胞形态的变化和DNA片段的变化。薯蓣皂苷元及相关衍生物能对Bcap37细胞产生抑制增殖的作用,并能诱导使之发生细胞凋亡。

       

      Abstract: The paper transformed diosgenin to a series of new analogues using Streptomyces virginiae IBL-14. The effects of diosgenin and its analogues on Hela cells, Bcap37 cells, KB-3-1 cells and SW1990 cells were tested by MTT method.The morphological changes of the Bcap37 cells treated with diosgenin and its analogues were observed micoroscopically by Hoechst 33258 staining. The apoptosis of the Bcap37 cells were evaluated by flow cytometry analysis. Diosgenin and its three analogues have inhibiting effects on the growth of the four tumor cells. The inhibiting effects on Bcap37 cells were better than the other three cell lines under the effects of diosgenin and its three analogues. Bcap37 cells showed typical characteristic of apoptosis, including the formation of apoptotic bodies. Diosgenin and its three analogues have evident inhibiting effects on the growth of Bcap37 cells and can induce apoptosis in Bcap37 cells in vitro. d
