    一种测定动物细胞平均沉降速率的新方法

    A Novel Method to Determine the Average Settling Velocity of Animal Cells

    • 摘要: 通过构建细胞沉降过程的理想化模型,提出了一种简易的细胞沉降速率测定方法。标准颗粒沉降实验结果验证了这种方法的准确性。利用该方法测定杂交瘤细胞的沉降速率,定量描述了杂交瘤活细胞与死细胞之间的沉降速率差异,活细胞的沉降速率(15.5mm/h)大约是死细胞(9.36mm/h)的1.67倍,约是细胞碎片(1.46mm/h)的10倍;把该方法用于不同种类不同培养时期的脐血造血细胞,发现它们具有不同的沉降速率。这些数据将为细胞连续灌注培养过程的细胞截留研究提供依据。

       

      Abstract: Perfusion has been verified as one of efficient and economical choice for animal cell culture bioprocesses. Cell retention within the perfusion system determines the efficiency of the whole processes to a great extent. While the settling velocity of animal cells plays an important role in the design of the cell retention device and the estimation of settling operation, the measurement of this critical parameter suffers from the lack of simple and accurate method for a long time. In this work a novel method was developed from a simplified mathematical model, and then validated with a well-design settling experiment using standard micro-particles. Based on the measurement of the settling velocities of the hybridoma and hematopoietic cells, this method provides a series of valuable data that may be potential for the subsequent cell retention investigations.
