    • 摘要: 使用生物信息学手段,从人公共蛋白质数据库的蛋白质序列中筛选到新的分泌蛋白基因CHID1(Chitinase domain containing 1),该基因定位于人11号染色体短臂15区5带(11p15.5),cDNA长1 182 bp,其编码蛋白共有393个氨基酸。转染COS-7细胞后Western blot实验显示:在细胞培养液中没有检测到CHID1蛋白。对CHID1基因在mRNA水平上进行组织分布考察发现:它在肺、肾、肝、心、胸腺中都有表达,肝中最高。细胞生长曲线揭示,Lovo-CHID1稳定细胞株有生长抑制的现象,而细胞周期分析发现细胞株在G1期发生阻滞。这表明CHID1有可能是个潜在的抑癌基因。

       

      Abstract: A set of bioinformatics methods were used to screen novel human secretory proteins from many protein sequences obtained from the public protein database in this study.A novel secretory protein gene,CHID1 was obtained.It locates in human chromosome 11p15.5,spanning 1 182 bp and encoding 393 amino acid residues.Transfecting in COS-7 cell and subsequently western blot analyzing showed that CHID1 was not detected in cell culture media.Tissue RT-PCR experiment uncovered that CHID1 was(expressed) in lung,kidney,l...
