    Effect of Nodosin Perfusion on the Expression of HO-1 in Hepar Tissue of SD Rat

    • 摘要: 探讨中草药溪黄草有效成分Nodosin对离体SD大鼠肝脏血红素加氧酶-1(HO-1)表达的影响。采用质量浓度为10 μg/mL的Nodosin溶液灌注离体SD大鼠肝脏,以乳酸钠林格氏液灌注作为对照,用RT-PCR和Western bloting方法观察两组离体肝脏组织HO1的蛋白和mRNA表达的变化。实验结果表明:经Nodosin溶液和乳酸钠林格氏溶液灌注保留1 h和2 h,肝脏组织中HO-1在蛋白和mRNA水平上表达较灌注完毕时均有上调(P<0.01),且实验组较对照组上调作用更加明显(P<0.01)。该结果说明溪黄草有效成分Nodosin具有诱导上调离体肝脏组织HO-1表达,从而对大鼠肝移植后缺血再灌注损伤起到有效的保护作用。

       

      Abstract: To examine the effect of nodosin extracted from Isodon serra on heme oxygenase-1(HO-1) in SD rat hepar in vitro,the isolated liver was perfused by Nodosin solution (10 μg/mL), while lactic Ringer's solution was used as control. Changes in the expression of HO-1 both in the mRNA and protein at 0, 1,2 h after perfusion by Western bloting and RT-PCR were examined. Results show that the expression of HO-1 both in the mRNA and protein is higher in perfused group than that in control,suggesting that Nodosin could induce HO-1 overexpression in isolated SD rat liver and protect hepar tissue by alleviating ischemia incurred by reperfusion.
