    聚乙二醇改性壳聚糖固定化L-天门冬酰胺酶反应性质研究

    Reaction Properties of L-Asparaginase Immobilized on Chitosan Modified with Polyethylene Glycol

    • 摘要: 研究了聚乙二醇改性壳聚糖(PEG-CS)固定化L-天门冬酰胺酶催化缓冲液中天冬酰胺的反应过程,考察了底物浓度、给酶量、温度、搅拌速率等条件对固定化酶在缓冲液中间歇催化反应过程的影响;以及底物浓度、流量、温度、床层高度等因素对固定化酶在缓冲液中连续催化反应过程的影响。实验结果表明:在适当的反应条件下,聚乙二醇改性壳聚糖固定化L-天门冬酰胺酶可以很好地水解缓冲液中的天冬酰胺。

       

      Abstract: Reaction properties of L-asparaginase immobilized on chitosan modified with polyethylene glycol were studied in this paper. The effects of substrate concentration, quantity of immobilized L-asparaginase, temperature, rotation rate on the batch reaction were investigated. Effects of substrate concentration, volume rate, temperature, height of enzyme column on the continuous reaction were investigated. The results show that under the proper conditions the immobilized L-asparaginase has good properties for hydrating asparagine from buffer.
