    Promotion by Microwave Radiation in the Extraction of Mahonia bealei Leaves

    • 摘要: 研究了在间歇式微波辅助萃取装置中提取阔叶十大功劳叶中的药用有效成分小檗碱。确定了萃取液中盐酸小檗碱的分析方法--Al2O3柱层析-紫外分光光度法;分别选取了甲醇、乙醇、乙醇-水(V/V=9:1)为溶剂;微波辐射时间为2min、4min、6min;处理比(g原料/mL溶剂)为1:40、1:60、1:80;利用正交实验设计考察了不同湿分原料的微波辅助萃取效率,得出了最优工艺条件;拍摄了典型萃取条件下萃取基本的透射电镜照片;探讨了微波辐射在植物原料萃取过程中对细胞结构破坏的机理。

       

      Abstract: A study on microwave assisted extraction (MAE) for berberine in Mahonia bealei was carried out in batch equipment. The mechanism of the extraction related to structural changes in the glands was discussed. The extracts were analyzed at 347nm by ultraviolet visible spectrophotometry. The parameters such as the solvent types, the intensity of microwave energy, the ratio (g/mL) of material to solvent volume were studied. The efficiency of microwave assisted extraction of different humidity material was investigated and optimized by means of the three factor and three level factorial design. Electron micrographs showed the mechanism of the extractions was related to structural change of the cells.
