Abstract:
A feasible assay method to determine brucine and strychnine in rat plasma by high performance liquid chromatography (HPLC) was developed. The pharmacokinetics of yaotongning capsule after intragastric administration in rats was studied. Results showed that the recovery of brucine and strychnine in different quality control concentrations surpassed 60%, intra day and inter day RSD (Relative Standard Deviation) were lower than 15%, stability and linearity were fine enough to meet the requirements of bioanalytical method. The kinetic processes of brucine and strychnine were all in accordance with two compartment open model after intragastric administration yaotongning capsule; Brucine assimilated slower, eliminated faster and had inferior bioavailability than strychnine. The method is convenient and reliable. It has good repeatability, precision, and stability. It can be applied to study the pharmacokinetics of yaotongning capsule.
