    High Density Suspension Cultivation of Taxus chinensis Cells

    • 摘要: 采用30g/L起始糖浓度,通过在细胞对数生长后期补料的方法成功地在250mL摇瓶和1.5L搅拌式生物反应器中进行了细胞高密度培养,经20d培养,细胞干重超过25g/L。在反应器培养中,次生代谢产物紫杉烷的产量较低。通过高密度培养,明显增加了细胞量,还可以提高反应器中紫杉烷的产量。

       

      Abstract: This paper studied high density suspension cultures of Taxus chinensis cells in both a 250 mL shake flask and a 1.5 L stirred bioreactor. In both cultivation vessels, a cell concentration of more than 25g/L was reached after 20 days of cultivation by sucrose feeding in the later stage of exponential growth phase. The production of taxane diterpene (taxuyunnanine C) was quite low in a batch bioreactor cultivation. Hight density cultivation could evidently increase the cell concentration and also enhance the production of the taxane in the bioreactor.
