    用双水相体系分离血红蛋白(HB)

    Separation of Hemoglobin by Aqueous TwoPhase System

    • 摘要: 利用聚乙二醇(PEG)/硫酸铵双水相系统分离纯化盐析得到了血红蛋白(HB)样品,考察了PEG相对分子质量、PEG质量分数、 (NH4)2SO4质量分数、pH、温度等因素对HB分离的影响。结果表明PEG双水相系统的最佳分离条件为:wPEG400=20%,w(NH4) 2SO4=20%,pH=7,温度为30 ℃。在此条件下HB分配系数可达到65,回收率可以达到89.5%,纯度达到80.5%。

       

      Abstract: The aqueous two-phase system of PEG/(NH4)2SO4, was used to purify hemoglobin(HB) in the initial salting out sample. The research investigated some parameters such as molecular weight of PEG, mass fraction of PEG and (NH4)2SO4, pH, temperature which will influence partition ratio of hemoglobin. The results show that the best separation condition: wPEG400=20%, w(NH4)2SO4=20%, pH=7, temperature 20 ℃. The highest partition ratio can reach up to 65, the maximum value of extractive rate was 89.5%, and its purity was 80.5%.
