    Study on covalent modification of superoxide dismutase and properties as well as metabolism of its modified form

    • 摘要: 右旋糖酐用高碘酸钠氧化或溴化氰活化后,与SOD共价结合制得Dx(IO_4)-SOD和Dx(CN)-SOD(Ⅰ)。它们的活力保存分别为81%和70%。在抗热、抗酸碱和抗蛋白水解酶等能力均比天然SOD高。静脉注射天然SOD和Ⅰ,在小白鼠血浆中的半衰期分别为4.06和41.46分钟,Ⅰ的半衰期明显提高,清除率大大下降。

       

      Abstract: Method for chemical Modification of superoxide dismutase by activation of dextran T70 with sodium periodate or cynogen bromide and subsequent coupling with superoxide dismutase is reported. The modified superoxide dismutases thus prepared rerains 81% and 70% of its original superoxide dismutase activity respectively.The dextran derivatives show higher resistance to pepsin and trypsin degradation, and their heat and pH stabilities are enhanced.Intravenous injection experiments prove that the present modification method is effective in increasing the half life of superoxide dismutase in mice plasma. It increases from 4.06 min to 41.46 min.
