    rhBMP-2对再生障碍性贫血小鼠骨髓造血恢复的促进作用

    Promoting Effects of rhBMP-2 on Aplastic Anemia Mice Bone Marrow Hematopoietic Recovery

    • 摘要: 探究了人重组骨形态发生蛋白-2(rhBMP-2)对再障小鼠的治疗作用。采用5-FU联合白消安建立小鼠再障模型,通过干预rhBMP-2进行治疗。考察了各实验组小鼠白细胞数、骨髓单核细胞数、体重、存活率、脾系数、粒系-巨系细胞集落(CFU-GM)数以及骨髓单核细胞中CD34+细胞比例,进行股骨、脾脏HE切片分析。结果表明,相较于再障对照组,rhBMP-2治疗组小鼠的白细胞数、存活率、CFU-GM集落数及骨髓单核细胞数显著提高,并且骨髓单核细胞中的CD34+细胞含量也提高,显著改善脾脏功能和缓解骨髓抑制,证实rhBMP-2能够促进再障小鼠造血损伤的修复。

       

      Abstract: Aiming to explore the promoting effects of rhBMP-2 on aplastic anemia mice bone marrow hematopoietic recovery,we built the hematopoietic injury model by 5-FU and busulfan with the interventional treatment of rhBMP-2.We analysed the number of mice peripheral white blood cells,bone marrow mononuclear cells,BM CD34+ cells,weight index,survival rate,spleen coefficient,CFU-GM colony as well as the HE sections of femur and spleen tissue.The results showed that rhBMP-2 can improve the index of peripheral white blood cell amount,mice survival rates,CFU-GM colony number,the bone marrow mononuclear cells number and CD34+ cells ratio;rhBMP-2 can also improve the function of the spleen and reduce bone marrow suppression compared with the AA control group.All results above confirmed that rhBMP-2 can effectively promote the restoration of hematopoietic injury in aplastic anemia mice.
